Project summary: The project aims to capitalize the renewable resurses by investigating some bacterial strains originating from zones with heterogenous environmental conditions where the differentiation of strains that are very productive for poly-β-hydroxybutyrates is high. This project is based on a great number of analyses required for highlighting the PHB synthesis capacity, namely microbiological analyses (estimating the soil microbial activity, isolating strong bacterial strains, assesing the physiology of PHB producing bacteria), molecular analyses (PCR amplification, sequencing, species identification, clonning, gene transfer from bacteria to yeasts, cell electroporation), biochemical (spectrophotometry), optical and electronical microscopy studies (TEM and SEM), culture physiology studies and biostatistical interpretations. The theme is scientifically justified both by the originality of the approach and by the attempt to understand the relation between the PHB bacterial producing capacity in different conditions, environmental and experimental ones. The applicability of the chosen theme presents multiple valences in environmental issues, biotechnology and medical field. Thus, the research project aimes to establish a protocol which to be used in a future biotechnology for obtaining poli-β-hydroxybutyrates.

The main goal of the project: The goals of this project are to isolate and characterize high poly-β-hydroxybutyrates PHB producing bacterial strains, to establish a protocol of the transfer of the gene involve in PHB synthesis from bacterial cells to yeast and to establish a biotechnological procedure for obtaining high levels of PHB. The strains will be isolated from different soil types in order to identify the environmental conditions which induce high levels of PHB.


1. Isolation of microbial strains from soils originating from physicochemically heterogenous zones.

2. The molecular identification of PHB producing species.

3. The parameters for effective PHB production will be optimised.

4. The transpher of the PHB producing-coding gene from bacteria to yeasts by means of a vector will be attempted.Tip proiect: PN II PD 393/2010

Rezumat: Proiectul isi propune valorificarea resurselor regenerabile prin investigarea unor tulpini bacteriene provenite din zone cu conditii de mediu heterogene unde probabilitatea diferentierii de tulpini inalt producatoare de poli-β-hidroxibutirati (PHB) este ridicata. Proiectul este bazat pe un numar mare de analize necesare pentru evidentirea capacitatii de sintetiza a PHB si anume analize microbiologice (estimarea activitatii microbiene din sol, izolare de tulpini bacteriene viguroase, fiziologia bacteriilor producatoare de PHB), moleculare (amplificare PCR, secventare, identificare de specii, clonare, transfer de gene de la bacterii la drojdii, electroporarea celulelor), biochimice (spectrofotometrie) studii de microscopie optica si electronica (TEM si SEM), studii de fiziologie a culturilor si intrepretari biostatistice. Tema este justificata stiintific atat prin originalitatea unui astfel de studiu cat si prin incercarea cunoasterii relatiei dintre capacitatea de producere de PHB de catre bacterii in diferite conditii de mediu cat si experimentale. Aplicabilitatea temei alese prezinta valente multiple in aspecte de mediu, biotehnologie si in domeniul medical. Astfel, proiectul de cercetare isi propune punerea la punct a unui protocol care sa fie utilizat intr-o viitoare biotehnologie de obtinere a poli-β-hidroxibutiratilor.

Scopul: Scopul acestui proiect de cercetare consta in izolarea si caracterizarea unor tulpini bacteriene cu capacitate ridicata de sinteza a poli-β-hidroxibutiratilor (PHB), in stabilirea unui protocol de transfer a genei responsabile de sinteza PHB de la bacterii la drojdii, precum si stabilirea unei proceduri biotehnologice de obtinere eficienta de PHB. Tulpinile bacteriene vor fi izolate din diferite tipuri de sol cu scopul de a gasi conditiile specifice care induc producerea unui nivel ridicat de PHB.


1. Izolarea tulpinilor microbiene din soluri provenite din zone cu conditii fizico-chimice heterogene.

2. Identificarea moleculara a speciilor producatoare de PHB.

3. Optimizarea parametrilor pentru producerea efectiva de PHB.

4. Transferul genei raspunzatoare de producerea de PHB de la specii bacteriene la drojdii prin intermediul unui vector.
